海洋酵母菌胞外酶及其基因的最新研究进展   总被引:4,自引:1,他引:3  
综述海洋酵母菌所产多种胞外酶的生产、纯酶性质及基因克隆工作的最新研究进展.从实验室建立的500多株海洋酵母菌种资源库中筛选出了数十株可以产生多种不同活性胞外酶的海洋酵母菌,它们分别属于耶鲁维亚酵母菌(Yarrowia)、类酵母(Aureobasidium)、毕赤氏酵母菌(Pichia)、梅奇酵母菌(Metschnikowia)和隐球酵母菌(Cryptococcus)等不同种属.所产生的胞外酶主要包括纤维素酶、碱性蛋白酶、酸性蛋白酶、淀粉酶(包括生淀粉酶)、菊糖酶、脂酶、植酸酶和嗜杀因子等.深入进行了各种胞外酶的生产,纯化及纯酶的性质研究,并对部分菌株的胞外酶基因进行了克隆、测序和表达,这些研究有助于对海洋酵母胞外酶的理论研究和生产应用价值的了解和对于海洋酵母菌资源、生理和遗传的进一步深入研究.  相似文献

A total of 328 yeast strains from seawater, sediments, mud of saltems, the guts of marine fish and marine algae were obtained. The results of routine identification and molecular methods show that five yeast strains obtained in this study belonged to Pichia spp., including Pichia guilliermondii luv-small, Pichia ohmeri YF04d, Pichiafermentans YF12b, Pichia burtonii YF11A and Pichia anomala YF07b. Further studies revealed that Pichia anomala YF07b could produce killer toxin against pathogenic yeasts in crabs while Pichia guilliermondii luv-small could produce high activity of extracellular inulinase. It is advisable to test if Pichia ohmeri YF04d obtained in this study is related to central-venous-catheter-associated infection.  相似文献

Little is known about the effects of offshore energy installations on the marine environment, and further research could assist in minimizing environmental risks as well as in enhancing potential positive effects on the marine environment. While biofouling on marine energy conversion devices on one hand has the potential to be an engineering concern, these structures can also affect biodiversity by functioning as artificial reefs. The Lysekil Project is a test park for wave power located at the Swedish west coast. Here, buoys acting as point absorbers on the surface are connected to generators anchored on concrete foundations on the seabed. In this study we investigated the colonisation of foundations by invertebrates and fish, as well as fouling assemblages on buoys. We examined the influence of surface orientation of the wave power foundations on epibenthic colonisation, and made observations of habitat use by fish and crustaceans during three years of submergence. We also examined fouling assemblages on buoys and calculated the effects of biofouling on the energy absorption of the wave power buoys. On foundations we demonstrated a succession in colonisation over time with a higher degree of coverage on vertical surfaces. Buoys were dominated by the blue mussel Mytilus edulis. Calculations indicated that biofouling have no significant effect in the energy absorption on a buoy working as a point absorber. This study is the first structured investigation on marine organisms associated with wave power devices.  相似文献

Sidescan sonar has been applied in a number of shallow water environments along the Dutch coast to map and monitor shellfish and seabed habitats. The littoral setting of these surveys may hamper data acquisition flying the towfish in zones of turbulence and waves, but also offers valuable opportunities for understanding, interpreting and validating sidescan sonar images because of the ability to ground-truth during low water periods, enabling easy identification and validation. Acoustical images of some of the mussel banks on the tidal flats of the Wadden Sea, recorded at high tide, show a marked resemblance with optical Google Earth images of the same banks. These sonar images may thus serve as ‘acoustic type signatures’ for the interpretation of sonar patterns recorded in deeper water where ground-truthing is more difficult and more expensive. Similarly, acoustic type signatures of (Japanese) oyster banks were obtained in the estuaries in the southwest of the Netherlands.  相似文献

在浙江近岸海域采集了青蛤(Cyclina sinensis)、毛蚶(Scapharca subcrenata)、厚壳贻贝(Mytilus coruscus)、缢蛏(Sinonovacula constricta)、泥螺(Bullacta exarata)、菲律宾蛤仔(Ruditapes philippi-narum)、文蛤(Meretrix meretrix)、僧帽牡蛎(Ostrea cucullata)和栉孔扇贝(Chlamys farreri)共9种贝类计29份样品,检测其中重金属汞、砷、铜、铅、锌、镉以及麻痹性贝毒和腹泻性贝毒.结果表明,瑞安毛蚶、瑞安栉孔扇贝中有麻痹性贝类毒素检出,嵊泗文蛤、毛蚶和乐清牡蛎中存在重金属超标的情况;牡蛎对汞、铜、锌、镉有较强的富集能力.  相似文献

通过检测深圳沿岸海域7种贝类的重金属、麻痹性贝毒(PSP)和腹泻性贝毒(DSP)含量,了解其食用安全性.严格按照国家标准进行检测,采用SPSS 20.0对数据进行Kruskal-Wallis H检验和方差分析.结果表明:各种重金属和贝类毒素在样品中均有检出,汞污染状况良好;铜含量范围为0.52~109.65 mg/kg,超标率为13.67%;铅含量范围为0.01~3.60 mg/kg,超标率为68.35%;镉含量范围为0.015~7.807 mg/kg,超标率为53.96%.PSP和DSP检出率为100%,PSP含量范围为0.30~34.23 ug/100 g;DSP含量范围为1.88~38.41 ug/100 g.总体看来,样品铅、镉超标严重,不同种类贝类重金属含量具有差异且贝类毒素检出率较高,应引起有关部门的重视,加强该海域监测工作.  相似文献

Green-lipped mussels (Perna viridis) were collected from seven fish culture zones (FCZs) in Hong Kong and analyzed for okadaic acid (OA). A conventional HPLC method was modified by incorporating a proteinase K digestion step. Results suggest that a higher recovery (2.5 times higher) of OA was obtained from spiked samples after the addition of 1.08 mg proteinase K in comparison with samples incubated without the proteolytic enzyme. For the hepatopancreas (HP) of individual field-collected mussels, the additional digestion step can enhance OA extraction by 3.1 times. Spatial and temporal variations in OA concentrations in the mussels from various FCZs were investigated. The highest concentration of OA in mussel HP samples was 1164.9 ng/g HP wet wt. With respect to OA concentrations in whole mussel tissues from seven sites and four seasonal samplings, the concentrations were between 70.0 and 131.0 ng/g wet wt., which did not exceed the generally recognized international regulatory criteria (>200 ng/g) for OA.  相似文献

Blooms of the toxic dinoflagellate Alexandrium fundyense commonly occur in the western Gulf of Maine but the amount of toxin observed in coastal shellfish is highly variable. In this study, a coupled physical–biological model is used to investigate the dynamics underlying the observed A. fundyense abundance and shellfish toxicity in 1993 (a high toxicity year) and 1994 (low toxicity year). The physical model simulates the spring circulation, while the biological model estimates the germination and population dynamics of A. fundyense based on laboratory and field data. The model captures the large-scale aspects of the initiation and development of A. fundyense blooms during both years, but small-scale patchiness and the dynamics of bloom termination remain problematic. In both cases, the germination of resting cysts accounts for the magnitude of A. fundyense populations early in the spring. Simulations with low net A. fundyense growth rates capture the mean observed concentration during the bloom peak, which is of similar magnitude during both years. There is little evidence that large-scale changes in biological dynamics between 1993 and 1994 were a primary driver of the differences in shellfish toxicity. Results instead suggest that the persistent southwesterly flow of the western Maine Coastal Current led to A. fundyense populations of similar alongshore extent by late May of both years. This period coincides with peak cell abundance in the region. Variations in wind forcing (downwelling favorable in 1993, upwelling favorable in 1994) and subsequent cell transport (inshore in 1993, offshore in 1994) in early June then provides a plausible explanation for the dramatic mid-June differences in shellfish toxicity throughout the western Gulf of Maine.  相似文献

为探讨底物中钙赋存形态对牡蛎幼虫附着的诱导效应,本研究设计3因子随机区组实验,检验实验底物钙含量、钙赋存形态及海水中牡蛎幼虫丰度对实验底物上太平洋牡蛎(Crassostrea gigas)稚贝附着效果(密度和壳高)的影响。结果表明,在3个因子中,实验底物钙含量对附着稚贝密度和壳高的影响不显著(P0.05),而其它2个因子(钙赋存形态和海水中牡蛎幼虫丰度)均显著影响着实验底物上附着牡蛎稚贝的密度和壳高(P0.05)。在相同的牡蛎浮游幼虫丰度下,3种钙形态底物对牡蛎幼虫附着的诱导能力大小顺序为:有机钙碳酸钙=硫酸钙(P0.05);壳高的结果却恰恰相反。在同样的钙形态底物中,高牡蛎幼虫丰度下牡蛎稚贝附着量均显著高于低幼虫丰度的处理组(P0.05)。  相似文献
